| Identification of a potential drug for the treatment of post-menopausal osteoporosis and fracture healing from our group has recently led to the development of new medication for bone health. We have standardized a fraction from Dalbergia sissoo or Sheesham as an osteogenic agent that has rapid fracture healing efficacyand will also be valuable in primary osteoporosis. In this standardized extract have identified a novel isoflavonoidCaviunin 7-O-[b-Dapiofuranosyl-(1-6)-b-D-glucopyranoside] abbreviated as CAFG that is present in the highest percentage.CAFG administration in rodents improved trabecular microarchitecture of the long bones and increased strength at the vertebral and femur sites. CAFG is also an effective accelerant for chondrogenesis and has stimulatory effect on the repair of cortical bone as observed after drill-hole injury at tissue and cellular level. Signal transduction inhibitors in osteoblast demonstrated involvement of p-38 mitogen-activated protein kinase pathway that is stimulated by BMP2 to initiate Wnt/b-catenin to be involved in this process. Overall, CAFG inhibits phosphorylation of GSK3-β and subsequent nuclear accumulation of b-catenin.These osteogenic effects were confirmedby abrogation observed by Dkk1, Wnt-receptor blocker and FH535, inhibitor of TCF-complex by reduction in β-catenin levels.
