RESEARCH ACTIVITIES
My research group is mainly focused on two major topics of organic and medicinal chemistry as mentioned below:
(i) Drug Development of Biologically Important Natural and Synthetic Products

During drug development program on Reproductive health including management of osteoporosis and bone related disorders, CSIR-CDRI research group observed that crude extract of Butea monosperma exhibited in vitro and in vivo bone forming (osteoblast mineralization) activity (Pandey et al. Menopause, 2010, 17, 602-610). Fractionation-guided activity showed a mixture of five isoflavonoids having good osteogenic activity. In order to identify the activity of individual isoflavonoids, high quantities of medicarpin and cajanin were required. We developed efficient protocol for the synthesis of these natural isoflavonoids (medicarpin and cajanin) and their new derivatives, and demonstrated their differential actions in osteoblasts and parameters of acquisition of PBM in growing female rats (Tyagi et al. Molecular and Cellular Endocrinology 2010, 325, 101-109; J. Nutritional Biochemistry, 2012, 23, 27-38; Osteoporosis International, 2012, 23, 1151-1161). We further prepared a series of new pterocarpans and showed that they hold therapeutic promise in postmenopausal osteoporosis (Goel et al.: Substituted benzofurochromenes and related compounds for the prevention and treatment of bone related disorders:US-8686028 dated 01-04-2014).Several New leads were identified as potent osteogenic agents and one of the pterocarpans CDRI-1500 (Pteroheal) significantly stimulated callus formation and increased fracture healing at only 1.0 mg/kg/day dose in adult rats. New bone formation at the fracture site is increased by ~40% in rats treated with CDRI-1500 compared to vehicle treated rats. Pteroheal (CDRI-1500) is currently under development mode and it is found safe in regulatory toxicology and pharmacology studies. Pteroheal is now ready for Licensing and we are looking for industrial partner for further development.

 
 

(ii)New Organic Fluorescent Molecules through Ring Transformation Strategy and their Applications

In Biological Sciences

We have designed and synthesized Donor-Acceptor Fluoranthene Dye FLUN-550 for Selective Fluorescence Staining of cytoplasmic lipid droplets of 3T3-L1 pre-adipocytes as well as for selective staining of neutral lipid droplets of Leishmania donovani parasite and soil nematode C. elegans for live and fixed cell imaging applications in chemical and molecular biology (Goel et al. Patent Appl. No. 807DEL2013 dated 19/03/2013; Organic Letters, 2014, 16, 756-759). Very recently, our group has discovered and reported ‘First’ dual colorimetric-ratiometric fluorescent probe ‘NAP-3’ for selective and direct visualization of Labile Iron (III) pools in multicellular oragnaisms (Soil nematode C. elegans) (Goel et al. Chem. Commun. 2015, 51, 5001). This probe has opened new avenues for enhancing our understanding on the crucial role of iron-catalyzed generation of reactive oxygen species, which is associated with diseases like cancer, diabetes and neuro-degenerative disorders.

 
 

In Material Sciences

Our group is engaged in the development of New Organic Fluorescent Dyes with absorption and emission at variety of wavelength for biological (as drug candidates or biomarkers) and material (smart electroluminescent devices) applications. Recently we have demonstrated a novel ‘concept’ of inhibiting “Green Emission Defect” in fluorene-based OLEDs by transforming “green-yellow” fluorenone molecules to “blue” fluorenone compounds by appropriate positioning of donor-acceptor and chromophoric groups onto widely used fluorene/fluorenone -conjugated system (Goel et al. PCT patent WO/2009/122445 dated 08.10.2009; PCT/IN2009/000215 dated 31.03.09). The performance of a blue fluorene was investigated by fabricating a multilayer OLED with a configuration of ITO/PEDOT:PSS/TPD (30nm)/D/A-fluorene (60nm)/BCP (8nm)/ LiF(0.5nm)/Al (160 nm), which revealed stable bright blue emission with a low turn on voltage of 3.5V and a brightness of 573 Cd/m2 with good chemical, thermal and electrochemical stability (Goel et al. Organic Letters, 2009, 6, 1289). We have shown that emission color can be effectively controlled from ‘blue to green to yellow to red’ by modulating the aromatic -characteristic not only in solution (photoluminescence) but also in solid electroluminescent devices (Goel et al. J. Mater. Chem. 2012, 22,14880). We have successfully prepared (In collaboration with IIT Electrical Engineer Dr. RS Anand at Kanpur) Yellow Fluorescent ‘Panels’ for exploring their potential for technology development.
